fa عدم ارتباط چند شکلی‌‌های  7G-A  و 138C -T  ژن MGP با بیماری عروق کرونر در بیماران ایرانی <p dir="rtl" style="text-align: justify"><!--stripped--> Normal 0 false false false EN-GB X-NONE AR-SA MicrosoftInternetExplorer4 <!--stripped--><!--stripped--> <!--stripped--> <!--stripped--> <style> /* Style Definitions */ table.MsoNormalTable {mso-style-name:"Table Normal" mso-tstyle-rowband-size:0 mso-tstyle-colband-size:0 mso-style-noshow:yes mso-style-priority:99 mso-style-qformat:yes mso-style-parent:"" mso-padding-alt:0cm 5.4pt 0cm 5.4pt mso-para-margin:0cm mso-para-margin-bottom:.0001pt mso-pagination:widow-orphan font-size:11.0pt font-family:"Calibri","sans-serif" mso-ascii-font-family:Calibri mso-ascii-theme-font:minor-latin mso-fareast-font-family:"Times New Roman" mso-fareast-theme-font:minor-fareast mso-hansi-font-family:Calibri mso-hansi-theme-font:minor-latin mso-bidi-font-family:Arial mso-bidi-theme-font:minor-bidi} </style> <!--stripped--> <a name="OLE_LINK1"><strong>زمینه و هدف</strong></a>: بیماری عروق کرونر Coronary Artery Disease CAD از شایع‌ترین علل مرگ و میر در همه کشورها از جمله ایران می‌باشد. از عوامل مستعدکننده بیماری، سابقه خانوادگی مثبت است که اشاره به ماهیت ژنتیکی CAD دارد. شناخت فاکتورهای ژنتیکی مستعدکننده به CAD، برای شناسایی افراد در معرض خطر بسیار مهم است. تغییر در فاکتورهای محیطی مستعدکننده و احیانا تداخلات دارویی در این افراد، می‌تواند نقش اساسی در پیشگیری از بیماری داشته باشد. بررسی‌های ژنتیکی تاکنون نقش بیش از 100 ژن را در ابتلا به CAD نشان داده‌اند. ژن  Matrix Gla Protein (MGP) از جمله ژن‌هایی می‌باشد که نقش چندشکلی‌‌های آن در جایگاه‌های 7- و 138- در پرموتور ژن، در ایجاد خطر ابتلا به CAD در بعضی از جمعیت‌ها گزارش شده است. مطالعه حاضر به بررسی نقش چندشکلی‌های مذکور در ایجاد خطر ابتلا به CAD، در جمعیت ایرانی پرداخته است. <strong><br></strong></p><p dir="rtl" style="text-align: justify"><strong>روش بررسی</strong>: 150 فرد مبتلا و 150 شاهد مناسب بر اساس علائم کلینکی و نتایج آنژیوگرافی انتخاب و از نمونه‌های خون DNA استخراج گردید و ژنوتیپ نمونه‌ها در هر دو چندشکلی با روش Polymerase Chain Reaction-Restriction Fragment Length Polymorphism PCR-RFLP و راندن محصولات روی ژل پلی‌آکریل آمید مشخص گردید. <br></p><p dir="rtl" style="text-align: justify"><strong>یافته‌ها</strong>: مقایسه فراوانی ژنوتیپی و آللی بین گروه بیماران و شاهد نشان داد که آلل A در جایگاه 7- و آلل T در جایگاه 138- در بین بیماران بیشتر از گروه شاهد می‌باشد. هر چند این اختلاف از لحاظ آماری معنی‌دار نمی‌باشد. <strong><br></strong></p><p dir="rtl" style="text-align: justify"><strong>نتیجه‌گیری</strong>: نتایج حاصل پیشنهادکننده عدم همبستگی چند شکلی‌های مذکور با CAD در جمعیت ایرانی می‌باشد. تایید قطعی این امر نیازمند تکرار مطالعات مشابه می‌باشد.</p> <p style="text-align: justify"><!--stripped--> Normal 0 false false false EN-GB X-NONE AR-SA MicrosoftInternetExplorer4 <!--stripped--><!--stripped--> <!--stripped--> <!--stripped--> <style> /* Style Definitions */ table.MsoNormalTable {mso-style-name:"Table Normal" mso-tstyle-rowband-size:0 mso-tstyle-colband-size:0 mso-style-noshow:yes mso-style-priority:99 mso-style-qformat:yes mso-style-parent:"" mso-padding-alt:0cm 5.4pt 0cm 5.4pt mso-para-margin:0cm mso-para-margin-bottom:.0001pt mso-pagination:widow-orphan font-size:11.0pt font-family:"Calibri","sans-serif" mso-ascii-font-family:Calibri mso-ascii-theme-font:minor-latin mso-fareast-font-family:"Times New Roman" mso-fareast-theme-font:minor-fareast mso-hansi-font-family:Calibri mso-hansi-theme-font:minor-latin mso-bidi-font-family:Arial mso-bidi-theme-font:minor-bidi} </style> <!--stripped--> <strong>Background:</strong> Coronary Artery Disease (CAD) is a major cause of death worldwide including Iran.  The risk of developing disease in patients without symptoms is assessed in part by factors that are associated with disease. Among these factors family history points to the significance of genetic component in the risk of CAD. The identification of the genetic variants that confer risk for CAD is essential for detecting high-risk individuals, so preventative life style and therapeutic action can be taken before overt disease develops. So far more than 100 genes have been reported with possible role in developing risk for CAD. Matrix- Gla Protein (MGP) is one of these genes that association of its single nucleotide polymorphism (SNP) with CAD has been reported.  Among the polymorphisms, there are two promoter SNPs at position -7 & -138 that their association with CAD has been reported before. Here we investigated the association of these SNPs with CAD in Iranian population. <br><strong>Methods</strong><strong>:</strong> 150 cases and 150 controls were selected on the basis of their clinical assessments and angiographic reports. DNA was extracted from blood samples. The genotypes for both SNPs were determined using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) method with size fractionation on Polyacrylamide gel. <br> <strong>Results:</strong> The comparison of allele & genotype frequencies between patients and controls showed that there is an excess of A allele at position -7 and T allele at position -138 among patients, although these differences were not significant (p<0.2, and p<0.5 respectively).<br><strong>Conclusions:</strong> This study suggests no association of these SNPs with CAD in Iranian population.
